Iyer scored 105 off 90 balls
While Gill scored 104 off 97 balls.
The pair put on a dominant 200-run partnership for the second wicket.
Suryakumar Yadav also chipped in with an unbeaten 72 off 37 balls to boost India's total.
The victory is a major boost for India's confidence ahead of the ODI World Cup in India later this year.
